Saint John Orlov was born on 1 November 1881 in the village of Limbuchany, in the Moscow province. He was the son of Priest John Petrovich Orlov. In 1897, he graduated from the Perervinsky Ecclesiastical School, while in 1903 he completed his studies at the Moscow Theological Seminary, after which he was ordained to the priesthood. He was appointed to serve at the Church of the Holy Trinity in the village of Troitskoye, in the Zvenigorod district, near the town of Voskresensk.
By 1937, the clergy of the Church of the Holy Trinity consisted of Saint John, Father Alexander Mashkov and Deacon Peter Troitsky. On 7 November 1937, Father Alexander was arrested and forced to testify against his fellow clergymen. On 18 November 1937, Father John Orlov and Deacon Peter Troitsky were arrested and imprisoned in Taganka Prison.
During the interrogations, Father John steadfastly denied all the charges brought against him. On 27 November 1937, an NKVD troika sentenced him to ten years of imprisonment in a forced labour camp.
He reposed on 7 March 1938 in the Mariinsk forced labour camp, in the Kemerovo region, and was buried in an unmarked grave.
